Selectoprobe™ Horseradish Peroxidase (HRP) (High Purity, ≥250U/mg)
Catalog No.: SP-F-0159
Description: Selectoprobe™ Horseradish Peroxidase (HRP) is a high-purity horseradish peroxidase enzyme designed for optimal use in activity assays and antibody conjugation applications, including ELISA, Western blot, and immunohistochemistry.
Key Features of Selectoprobe™ Horseradish Peroxidase:
- Purified and Ready-to-Use: Lyophilized, salt-free powder form for easy dissolution and immediate use.
- High Specific Activity: Typically greater than 300 units/mg (with lot-specific activity values provided).
- Superior to AP: HRP (40kDa) is smaller and has a higher specific enzyme activity compared to alkaline phosphatase (AP; 140kDa) and beta-galactosidase (b-Gal).
- Versatile Applications: Compatible with numerous substrate solutions and assay techniques, providing flexibility in assay design.Supplied as a lyophilized, salt-free powder, this HRP preparation is ideal for molecular biology and protein research applications, particularly as a reporter system in immunoassays and probe-based methods like ELISA, Western blotting, EMSA, and Southern blotting. Typically conjugated to secondary antibodies or streptavidin, HRP activity can be visualized using colorimetric or luminescent substrates.
Horseradish peroxidase (HRP) interacts with hydrogen peroxide (H₂O₂) to form an [HRP-H₂O₂] complex, which can oxidize a broad range of hydrogen donors, including various substrates used in detection. These substrates include colorless, reduced chromogenic substrates (e.g., DAB, TMB, ABTS), chemiluminescent substrates (e.g., luminol), and fluorescent substrates (e.g., Amplex Red). Upon incubation with these substrates, HRP produces a colored, luminescent, or fluorescent product, allowing quantification through colorimetric, luminescent, or fluorimetric detection methods.
HRP can be conjugated to antibodies using several techniques, such as glutaraldehyde, periodate oxidation, disulfide linkage, or amino- and thiol-directed cross-linkers. Compared to β-galactosidase and alkaline phosphatase, HRP’s smaller size and enhanced stability make it a preferred labeling enzyme for many applications.
